Acetylated polypeptide chains of the four hemoglobin (Hb) components (F1, F2, S1, and S2) of eel, Anguilla japonica, were analyzed for N-terminal amino acid sequence by use of the method involving "Pronase" and carboxypeptidase digestions. Results obtained are summarized as follows: (1) The N-terminal structure of the acetylated polypeptide chain was Acetyl-Ser-Leu-Ser-Ala-… for component F1, and Acetyl-Ser-Glu-Ser-Asp-… for the other three components. (2) Principally based on a close resemblance in the N-terminal sequences of those acetylated chains of eel Hb's to the α chain of carp Hb, it could be concluded that the acetylated chains of the four Hb components correspond to the α chains of Hb's from human or other vertebrates, and hence the valyl chains of eel Hb's reported previously, to the non-α chains, or to the β chains as called customarily.
