HUBBARD'S benzidine colorimetric method for determining sulphuric acid was examined Following points must be borne in mind: -(1) Colorimetry must be done from 40 to 60 minutes after adding 8 cc of 0.1N ch'oric acid to 20 cc of sample solution. (2) Ratio between the concentration of the sample and that of standard solution must be'kept far within the limits of 2 and 1/2. (3) With the aid of the definite relation which exists between the true content and the detected amount, the latter must be corrected. (4) Precipitation of benzidine sulphuric acid must be progressed in the presence of trich'or-acetic acid. By the method stated above, sulphuric acid in agar-agar were detected to amount to 2.156% total sulphuric acid in SO4, without no free acid. Agar-agar used was not hydrolysed by TAKA-diastase. With carps, 0.0864% of inorganic sulphuric acid in SO4, were detected from muscle, 0 0328.mg per one cc from serum but none from the other organs examined, i. e. liver, pancreas, spleen, ovary, kidney and gall-bladder.
