Effects of ethanol (EtOH) and acetaldehyde (AA) on endocytosis of cultured rat Kupffer cells, prepared by pronase digestion and density gradient cetrifugation from Wistar male rats, were investigated with the use of light and electron microscopy. Cultured Kupffer cells treated with 50mM or 100mM of EtOH showed reduced number of cytoplasmic projections and depression of phagocytosis of 2.2μm latex particles, in spite of no depression of endocytosis of 0.59μm latex and OK-432 particles (streptococcal preparation). Cultured Kupffer cells treated with 200μM AA showed no remarkable changes of cytoplasmic projections or endocytosis of particles. These results suggest that EtOH directly decreases the number of cytoplasmic projections and inhibits the phagocytosis of cultured rat Kupffer cells.
