Mechanisms for transport and conjugation of bilirubin glucuronides in hepatocytes haven't been defined. To examine factors affecting relative concentrations of BMG and BDG in bile, 1 studied bilirubin metabolism using ordinary Wister rats and Wistar Kyoto strain rats (Wister Kyoto and SHR).
BDG content in bile of latter was significantly dominant and accounted for 80-90% of bilirubin metabolites, whereas that of Wistar rats accounted for 50-60%.
Microsomal formation of BMG and BDG with native microsomes revealed higher activity in the latter. The rate of BMG conversiton to BDG was dominant and BMG release from microsomes was restricted and BDG accumulated in microsomes.
Exogenous BMG was excreted in bile without further conjugation in the latter, while 23% of BMG was converted to BDG in the former.
These findings indicate differences of intrahepatic transport, activity of UDPGT and the membrane enviroment between them.
