To clarify the role of Kupffer cells in endotoxin-induced liver injury, modifications of liver injury administrations of immunopotentiators and a complement inhibitor were studied.<BR>E. coli 026: B6 endotoxin (Difco Lab.) was injected into 98 Sprague-Dawley strain rats two times at 48 hours interval. At 48 hours after the second endotoxin injection, all survival rats were sacrificed and serum liver function tests and histological examination of livers were performed.<BR>In comparison with non-treated control, groups treated with immunopotentiators [PSK (Krestin), lysozyme (Neuzym), levamisole, OK-432 (Picibanil)] and a complement inhibitor [K-76] showed slight degeneration of hepatic cells, marked proliferation of Kupffer cells and low levels of S-GOT and S-LDH. Fspecially lysozyme treatment was most effective in the administrations of various immunopotentiators.<BR>It is concluded that Kupffer cells play a very important role in protecting a liver from endotoxin- induced liver injury.
