ADCC of peripheral blood lymphocytes from thirteen normal subjects was investigated in a assay system of <SUP>51</SUP>Cr-labeled chicken red blood cells (CRBC) coated with rabbit antiCRBC serum. Results were as follows; 1) ADCC of unfractionated mononuclear cells was 66.1±9.5% (M±SD). 117±0.6% decrease of this activity was demonstrated following monocyte-depletion from the peripheral blood cells. 2) Maximal effect was obtained under condition of effector to target cell ratio of 25:1 and antiserum dilution of 10<SUP>-3</SUP>. 3) Investigation of time kinetics of ADCC revealed that target cell lysis progressed rapidly until 4 hours and reached to the maximal effect at 6 hours. This effect was perpetuated until 36 hours after the culture. 4) Fractionation of lymphocytes demonstrated that effector cells were non T-lymphocytes. 5) Pretreatment of lymphocytes with heat-aggregated IgG at concentration of 1-10mg/ml suppressed cytotoxicity completely. Slight suppression of the cytotoxicity was also found at concentration of 0.01mg/ml.
