Recently, the existence of estrogen receptor (ER) in the tissue of colorectal cancer has been reported. To investigate the clinico-pathological characteristics of receptor-positive primary, colorectal cancers, biochemical assay of ER and progesterone receptor (PgR) was carried out in 38 cases, fluorescence cytochemistry in 43 cases and ER staining with monoclonal anti-ER antibody (ER-ICA) in 44 cases. Staining of endogenous estradiol (E2) and progesterone (PRG) in 112 primary colorectal cancers was also performed using Vectastain ABC Kit (ABC method).<BR>The positive rates were 15.8% for cytosol ER, 14.7% for nuclear ER, 15.6% for cytosol PgR and 21.4% for nuclear PgR by biochemical assay, and 16.3% for ER and 18.6% for PgR by fluorescence cytochemistry, while no positive case could be detected by ER-ICA. The positive rates by ABC method were 23.2% for E2 and 19% for PRG.<BR>In most of the receptor-positive cases, the tumors were advanced and had infiltrated deeply into the wall of the colon and rectum. They showed a tendency to be ill-defined ulcerated cancer (Borrmann 3-like cancer), moderately differentiated adenocarcinoma, n(+), ly (+) and v (+).<BR>It seemed that fluorescence cytochemistry was better than the other methods for the detection of ER and PgR in colorectal cancers.<BR>The presence of ER and PgR in some colorectal cancers suggested the possibility that the hormones (E2 and PRG) are concerned with colorectal cancers. Thus, endocrine therapy might contribute to the treatment of colorectal cancers.
