Effects of Intravenous Injection of Hens with Salmonella Antigen on the Distribution of Major Histocompatibility Complex Class II Positive Cells in Ovarian Follicles

The goal of this study was to determine whether the distribution of major histocompatibility complex class II positive (MHC class II+) cells was affected by foreign agents in the blood circulation. White Leghorn layers were injected intravenously with or without formalin fixed Salmonella paratyphi, and the area of MHC class II+ cells in the ovarian follicles, which included the largest (F1) and third largest follicles (F3), white follicles (WF) and cortical follicles, was examined by immunocytochemistry. The immunoreaction products for MHC class II were observed in cells of the thecal layer of all follicles in both treated and control birds. Salmonella paratyphi antigen caused a significant increase in the area of MHC class II+ cells in the theca of the F1, F3 and WF, but not in the cortical follicles. The results suggest that the MHC class II+ cells in the theca increase in response to circulating foreign agents, and play a significant role in ovarian local immunity.本実験は、ニワトリの卵胞において免疫応答に重要な主要組織適合遺伝子複合体(MHC)クラスII陽性細胞が外来抗原に反応して変化するかどうかを明らかにすることを目的とした。白色レグホーン種産卵鶏にSalmonella paratyphi(SP)死菌または生理食塩水(対照区)を静脈内投与し、卵巣皮質卵胞、白色卵胞(WF)、第1位および3位卵胞(F3とF1)におけるMHCクラスII陽性細胞の分布を免疫組織学的に解析した。MHCクラスIIに対する免疫反応産物は、対照区とSP投与区のいずれにおいても、すべての卵胞の卵胞膜に認められた。SP投与により、WF, F3とF1の卵胞膜でMHCクラスII陽性細胞の分布は有意に増加したが、皮質卵胞ではこの増加は認められなかった。これらの結果から、卵胞膜においてMHCクラスII陽性細胞は外来抗原に応答して増加し、卵胞の局所免疫に重要な役割を果たすと考えられた。
