Peripheral Blood Stem Cell Collection in Accordance to Monitoring of CD34 Cells

In order to determine the timing of peripheral blood stem cell (PBSC) collection, we monitored the CD34 cells appearing in peripheral blood after mobilization. The PBSC were collected using an AS104. The CD34 cells were counted by flowcytometry. For mobilization, granulocyte-colony stimulating factor (G-CSF) was injected to patients from nadir of circulating white blood cells (WBC). When the WBC increased to 3-3.5 × 10^4/μl, the injection of G-CSF was discontinued. In most patients, the WBC reappeared within 5-10 days after the nadir, accompanied by an increasing number of CD34 cells. Howev-er, the recovery of WBC was not always linear to the increasing number of CD34 cells. In a few cases, the recovery of WBC was delayed. In such cases, the CD34 cells did not increase and PBSC collection became unsuccessful. When the circulating WBC became 2-3 × 10^4/μl and circulating CD34 cells increased more than 3% in the WBC, CD34 cells of more than 2 × 10^6/kg were generally collected by processing 10,000 ml of blood. In order to avoid insufficient PBSC collection, the monitoring of CD34 cells is useful.
