A lethal ccdB gene-based vector was developed for the construction of shotgun libraries in Escherichia coli. The mutated ccdB gene carries multiple cloning sites and the amber stop codon, which simultaneously allows for maintenance of the intact vector in supE^- strains and removal of uncut and self-ligated plasmids in supE^+ strains.
