Lipid peroxide levels and platelet aggregation in the kitten blood were measured to determine the relationship between retinopathy of prematurity and lipid peroxide, and inhibitory effect of vitamin E on platelet aggregation was studied. One hundred and sixty-nine kittens were divided into 4 groups. Group A kittens were given oxygen, and group B kittens were not; group C kittens were administered oxygen and tocopherol acetate, and group D kittens were given oxygen and a vehicle placebo of tocopherol acetate. Kittens were placed in an infant incubator, and oxygen administration (70±1%) was started 3 days after birth and continued for 48 h. Measurement of lipid peroxide levels and ADP-induced platelet aggregometry were performed on days 5, 7, 9, 14, and 21. The results were as follows: 1. In group A, lipid peroxide levels were higher than in group B on days 7, 9, and 14, and the highest on day 7. 2. In group A, platelet aggregation on days 5 and 7 was higher than in group B. 3. Lipid peroxide levels were lower in group C than in group D. 4. Tocopherol acetate inhibited platelet aggregation on days 14 and 21 in group C. From these results, it was indicated that the increased lipid peroxide caused by oxygen administration enhanced platelet aggregation and that the formation of thrombus was accelerated. Therefore, it was concluded that these conditions were one of the causes of retiopathy of prematurity. Vitamin E had an inhibitory effect on platelet aggregation by lowering lipid peroxide levels.
