One hundred and eight specimens of uterine cervix were examined by preparing the rapid frozen sections and staining them with acridine orange. Among them, histologic examination revealed 5 cases of atypical hyperplasia, 4 cases of carcinoma in situ, 50 cases of invasive carcinoma of the cervix. In case of invasive carcinoma was studied the relationship between the fluorescent findings of nucleic acid by acridine orange staining and the histological degree of malignancy by light microscope. 1) It was confirmed by the treatment of the ferment that green to yellow fluorescence of cell nucleus was due to desoxyribonucleic acid, and red fluorescence of cell cytoplasm was due to ribonucleic acid. 2) In normal cervical epithelium nucleus of basal cell fluoresced intense yellow and toward the superficial cell layer nucleus fluoresced green. Cytoplasm of basal and para basal cell showed intense red fluorescence and as the cell became more mature, cytoplasm showed reddish brown to green fluorescence. 3) Nuclei of atypical hyperplasia in many cell layers showed intense yellow fluorescence, especially in cases of carcinoma in situ, all cell nuclei showed much intenser yellow fluorescence. 4) In many cases of low degree of infiltrative proliferation, nucleus of carcinoma cell showed intense bright yellow fluorescence and cytoplasm showed low red fluorescence. However, of high degree of infiltrative proliferation, nucleus of carcinoma cell showed brilliant orange-yellow fluorescence and cytoplasm showed intense flaming red fluorescence. 5) Ultra -thin sections of carcinoma tissues were observed with electron microscope atout the basement membrane and the stromal cell, and this electronmicroscopic findings were compared with the histologic findings of light microscope. In case of low degree of infiltrative proliferation the basement membranes of carcinoma nests were well preserved, but some parts of the nests showed the small defects of the basement membrane and the cytoplasm of carcinoma cell was protruded into the stroma. The degenerative changes of the fibroblasts and collagen fibrils were observed in low degree. However, in case of high degree of infiltrative proliferation the defects of most parts of the basement membrane and the degenerative changes of the fibroblasts and collagen fibrils were observed in high degree.
