The differences of spindle discharges between tonic red muscles and phasic white muscles were recently investigated by Granit and Homma (1959). Whether there are any structural differences corresponding to that of function in muscle spindles or not is an interesting problem. In this paper the structure of the muscle spindles from phasic and tonic muscles was investigated with special attention to the nuclear bags. Preparations were taken from gastrocnemius and tibialis anterior muscle as phasic muscle and soleus as tonic one and stained by gold chloride method (Cole, 1946). Comparisons between phasic, and tonic muscles were done about the length and the diameter of the primary endings, the diameter of the sensory nerve terminal wraps around the nuclear bags, Group Ia fiber diameter near the ending and the number of spiral rings per unit length r (50μ) of the nuclear bags. The differences between cats and monkeys were shown clearly. The most significant result was, however, the differences of diameters of the sensory nerve terminal wraps around the nuclear bags between phasic and tonic muscles. The largest was that from tibialis anterior muscle (38μ for the cat and 32μ for the monkey), the medium size from gastrocnemius (30, 29) and the thinest from soleus (29, 26). This order was corresponding to that of the contraction speed, tibialis anterior being the fastest and soleus the slowest. From these facts we could expect that there may be the following differences 1) the diameter of the nerve terminal wraps around the nuclear bag 2) the diameter of the nuclear bag itself 3) the diameter of the nuclear chain region (this was supposed from the diameter of the primary ending and the diameter of its nuclear bag since the former was almost same in phasic and tonic muscles) In 1) and 2) phasic muscles have larger values and smaller in 3) than tonic one. Possible explanation of these results is that if the nuclear bag fibers are responsible for the phasic component of muscle spindle discharges and there are some correlation between muscle fiber diameter and its contraction speed, then the nuclear bag fibers of tibialis anterior muscles will be more phasic than those of soleus. About the difference in 3) the discussion could not be carried out without large errors, for these were extremely indirect values.
