(1) Polyvinylpyrrolidone (PVP) is capable of prolonging the duration of action of drugs. This action of PVP appeared to be related to its local concentration and to be due to slowing down of the transfer of drugs from the site of action to the blood. (2) The blood pressure, measured directly and indirectly in rabbits and cats to which adrenaline, acethylcholine or hexamethonium (C_6) had been given intramuscularly or intraarterially in combination with PVP, indicated that the action of these drugs was weakened by PVP but the duration of action was twice as long as the normal duration. This action of PVP was not observed in intravenous injections of these drugs. (3) The lethal doses of C_6 and d-tubocurarine, when administered intramuscularly or intraperitoneally in combination with PVP, became smaller when the concentration of PVP was high. When PVP and d-tubocurarine were administered at longer intervals, the greater number of deaths resulted. (4) The action of some drugs on the heart removed from the frog or on the ileum extirpated from the mouse was inhibited by PVP. (5) In indirect stimulation of nerve-muscle preparation of the frog, the action of C_6 was caused by PVP to be delayed in taking effects. (6) Uptake of P^<32> by various organs of the mouse following its intramuscular administration decreased when PVP was administered in combination with P^<32>. (7) When a mixture of histamine with PVP was developed on a filter paper, the use of higher concentrations of PVP prevented its ascension from taking place, and the spots to be occupied by histamine became uncertain and unpredictable. (8) The appearance of 1 % fluorescein in the cornea after it was injected into the thoracic lymph vesicle of the frog was delayed when PVP was given in combination with 1 % fluorescein. The dispersion of a pigment in the layer of agar-agar in a test tube was inhibited by PVP.
