Regarding the effect of ultraviolet irradiation on antigen antibody reactions, the following findings have been obtained as a result of the experiments on precipitin reaction between normal human serum albumin and rabbit antiserum. Electric-photocell-nephelometer was used in the experiments. Although the first stage of antigen-antibody reactions, i.e. combining・of antibody with antigen, is completed, irradiation of antibody or antigen with a certain amount of ultra-violet rays causes change in the speed of flocculation (the process of aggregation of antigen-antibody complexes.) Both checkerboard experiments by means of layer method and supernatants test reveal results corresponding to those shown in test tubes in which the first stage of reactions has been completed. The results of experiments are summarized as follows: 1) Irradiation of antigen or antibody (contained in serum) retards flocculation. Multiplication of radiation time results in the variations in optimal ratio; when antigen is 瓶radiated the ratio varies in favor of antigen excess zone, but when antibody is irradiated it varies in favor of antibody excess zone. The amount of radiation being equal, ultraviolet rays have larger effect on antigen than on antibody, so far as these two variations are concerned. If radiation time is further multiplied 4〜8 fold, variations occur in the optimal ratio as determined by supernatants test as well as in the result of checkerboard experiments by means of layer method. In this instance also, pronounced change is observed when antigen is irradiated, whereas change is slight when antibody is irradiated. 2) As a matter of fact, various methods of test yield different results in titrations of antibody or antigen affected by ultraviolet irradiation. It will be seen, therefore, that the most rapidly flocculating mixture observed during test for combinations of antibody dilution and antigen dilution does not necessarily coincide with the optimal ratio. The author favors the observation of the point in checkerboard examinations where ultimate flocculation occurs by layer method.
