The following experiment was made to study the change in the quantity of adrenalin in the suprarenal capsule and in the blood which is brought about by the application of chloroform and ether. Method: mature female rabbits were used as material. The quantity of adrenalin in the capsule was examined by the Sudo-Inoue Method, a modification of Comesatti's and that in the blood by the Trendelenburg Method. As contrasts, some normal mature rabbits were used. Results: 1) The quantity of adrenalin in the capsule of the normal mature rabbits averaged 0.0964mgm. per 1kgm. of body weight. 2) When chloroform was applied to the animals, the following change in the quantity of adrenalin in the capsule was observable; 0.0707mgm. per 1kgm. of the body weight 30 minutes after the application of the anaesthetic, 1 hour later 0.0694mgm., 3 hours later 0.0866mgm., 6 hours later 0.0735mgm., 24 hours later 0.0643mgm., 48 hours later 0.0762mgm., 72 hours later 0.0922mgm., 96 hours later 0.1003mgm., and 144 hours later 0.0966mgm. The change in the quantity of adrenalin in the capsule, therefore, in the cases to which the anaesthetic was applied, was twofold, viz. the first decrease between 30 minutes and 1 hour after the application, and the secondary degrease 24 hours later. 3) When ether was administered, the quantity of adrenalin in the capsule calculated 30 minutes after the administration was 0.0770mgm., 1 hour later 0.0704mgm., 3 hours later 0.1036mgm., 6 hours later 0.1014mgm., and 24 hours later 0.0966mgm. In the cases of the application of ether, therefore, there was only one change, that which was observed between 30 minutes and 1 hour later. 4) In the cases to which chloroform was applied, the rate of decrease of the drops in the Trendelenburg preparation was observed to be 12.48% prior to the application, 30 minutes later 15.90%, 3 hours later 12.00%, 24 hours later 8.72% and 96 hours later 12.20%. In spite of the fact that quantity of adrenalin in the blood in the first period of decrease in the capsule was thus observed to be increasing, it decreased markedly in the second period of decrease. 5) In the cases to which ether was applied, the rate of decrease of the drops in the Trendelenburg preparation was observed to be 13.76% just prior to the application, 17.56% 30 minutes later, 15.16% 3 hours later, and 13.30% 24 hours later. The result was therefore the reverse of the above; an increase in the quantity of adrenalin in the blood was observed in the periods of decrease. 6) The quantity of adrenalin in the capsule of rabbits whose N. Splanchinicus had been cut off was also examined. When chloroform was administered to these rabbits, the results obtained were 0.1011mgm. in the first period of decrease viz. 1 hour after the application, 0.0772mgm. in the secondary period of decrease, viz. 24 hours later. In these cases, the decrease was shown only once, in the second period. When ether was applied, the quantity of adrenalin in the capsule was observed to be 0.1045mgm. in the first decreasing period, proving no decrease. According to the above results obtained, the first decrease in the quantity of adrenalin in the capsule brought about by chloroform is proved to be ascribable to the excessive consumption of adrenalin caused by the irritation of the central nerve. The same can be said with regard to the decrease in the case of ether. The secondary decrease of adrenalin brought about by chloroform should be ascribed to the decreased production of adrenalin caused by the obstacles in the capsule itself.
