In osseous tissue, osteoblasts form the extracellular matrix using type I collagen and non-collagenous proteins. Mineralized bone matrix is believed to result from calcium phosphate accumulating in matrix vesicles to be released from osteoblasts, then forming hydroxyapatite crystals that are deposited into collagen. Bone formation is regulated by various hormones, but assessing the effects of hormones on matrix mineralization has been difficult. The present study is the first to successfully and quantitatively assess matrix mineralization using SaOS-2 human osteoblastic sarcoma cells in vitro. This study also proved that mineralization is significantly elevated by incubating SaOS-2 cells with an appropriate concentration of hPTH in the presence of dexamethasone by histologically analyzing cells under scanning electronmicroscopy and biochemically analyzing cells using the 45Ca2+ radioactive isotope. These results will be very useful in the clinical application of hPTH for the treatment of steroid-induced osteoporosis in the future.
