Bone Conductive and Inductive Effects of Poly(L-Lactid and ε-Caprolactone) / Hydroxyapatite Composites and Bone Morphogenetic Protein-2 in Rats

Hydroxyapatite(HA)with copolymer of L-lactide andε-caprolactone[HA/P(LA/CL)]is expected to be a potent biomaterial for orthopedic surgeries. Bone morphogeneticprotein-2(BMP-2)has bone-inducing capability. In this in vivo study,weevaluated osteoconductivity(newly formed bone surrounding the implanted material)and osteoinductivity(newly formed bone in the implanted material)of HA/P(LA/CL)alone or combined with BMP-2 in cancellous bone in 7-month-old Sprague-Dawley rats.Surgical osteotomy in the sagittal orientation was performed on the proximal tibia andHA/P(LA/CL)alone or combined with BMP-2(1 or 10μg/block)was implanted tocontact with the osteotomized cancellous bone surface. Four and eight weeks postsurgery,bone specimens at the operated sites were harvested and osteoconductivity andosteoinductivity were evaluated with quantitative histomorphometrical method. HA/P(LA/CL)block alone showed osteoconductivity from the week four but did not showosteoinductivity. Combination of HA/P(LA/CL) block and BMP-2 showed bothosteoconductivity and osteoinductivity in a dose and time dependent manner. Becauseof its strong osteoconductivity and osteoinductivity,combination of HA/P(LA/CL)andBMP-2 might have potential clinical applications.
