RFP-トランスジェニッククローンネコの臍帯血由来間葉系細胞からの神経様細胞への分化(実験動物学)

臍帯血由来間葉系細胞は多能性のある成熟幹細胞で,間質組織や神経系列に分化することができる.本研究では赤色蛍光タンパク質(RFP)-トランスジェニックネコの臍帯血由来間葉系細胞の神経への分化能力について調べた.間葉系細胞はプレインキュベーションメディウムで24時間培養後,神経誘導メディウムで72時間培養した.免疫蛍光染色の結果, 6.85%の細胞がβIII-チュブリン陽性, 3.37%が神経フィラメント-Lに陽性, 7.04%が神経フィラメント-Mに陽性であった.βIII-チュブリンのバンドはウェスタンブロットでも検出された.これらの結果は, RFP-トランスジェニック臍帯血由来間葉系細胞がインビトロで神経細胞に分化できることを示している.それ故, RFP-トランスジェニック間葉系細胞は幹細胞移植において新たな追跡材料となる可能性を備えている.
