Despite many modern methods that are available for obtaining simple amino acids in their chiral form, it is not easy to obtain amino acids from carbon dioxide (CO_2), particularly in an efficient scalable fashion. In our process research for labeled amino acids, we focused on the synthesis of phenylalanine from carbon dioxide. Considering its reliability and versatility, we envisaged a general synthetic route from two important intermediates: cinnamic acid (A) and dehydroamino acid (B) as shown below. A robust synthetic route utilizing acetylene carboxylation and Trost's α-amination as key steps is summarized below (Scheme 1). Besides these important steps investigated, we have made intensive survey on the asymmetric hydrogenation of the phthalimide-protected dehydroamino acid, which is summarized in the Tables (shown in the previous pages). These investigations made it possible to prepare phenylalanine from carbon dioxide efficiently.
