In the course of screening for 1, 3-β-glucan synthase inhibitors, F-14232 C (1) and the related compounds were isolated from fermentation broth of a fungal strain SANK 17397. The main component, F-14232 C (1), strongly inhibited 1,3-β-glucan synthase from A. fumigatus and from C. albicans with IC_<50> values of 0.015 and 0.15μg/ml, respectively. In this reports we describe the structural elucidation of 1. The molecular formula of 1 was established to be C_<59>H_<105>N_<13>O_<18> by high-resolution FAB-MS. The IR and NMR spectra showed that 1 was a member of peptide antibiotic consisting of ten amino acids. Identification of each amino acid was achieved as follows. Conventional amino acid analyses along with NMR studies showed that 1 contains Asn (2 moles), Thr (2 moles), Ala (1 mole), Gly (1 mole) and β-Ala (1 mole). The γ-OH-Gln moiety was also revealed by NMR analyses. Furthermore, two uncommon amino acid residues, bearing long alkyl chains with a hydroxyl group were determined by MS spectral analyses. The arrengement of amino acids was elucidated by ROESY and HMBC spectra as shown in Fig. 4. Absolute configuration of amino acids was mainly determined by advanced Marfey's method. Based on the results, the structure of 1 was established to be a novel cyclic peptide as shown in Fig. 1.
