Ipecac, the roots of Cephaelis acuminata or C. ipecacuanha (Rubiaceae), is one of the most important medicinals from natural sources. Because of the emetic activity of its main alkaloids, emetine (1) and cephaeline (2), ipecac syrup is still indicated as an emetic for emergency use in the treatment of drug overdosage, and in some case poisoning. Previous phytochemical studies resulted in the isolation of six ipecac alkaloids from C. ipecacuanha. On the other hand Alangium lamarckii (Alan-giaceae) used in Indian folk medicine has been found to be a rich source of ipecac and Alangium alkaloids. On account of our interest in the biosynthesis of ipecac and Alangium alkaloids, we reexamined the alkaloidal constituents of C. acuminata and A. lamarckii and isolated seven new alkaloids. From the methanolic extract of the roots of C. acuminata, five novel alkaloids, neocephaeline (8), 7'-O-demethylcephaeline (9), 10-O-demethylcephaeline (10), 2'-N-(1"-deoxy-1"-β-D-fructopyranosyl)cephaeline (11), and 2'-N-(1"-deoxy-1"-β-D-fructopyranosyl)neocephaeline (12) were isolated along with 1-4, 6 and 7. From the methanolic extract of the fruits of A. lamarckii, two novel alkaloids 1', 2'-dehydrotubulosine (19) and 20 were isolated along with 2-6, 8, 10, 11, 14-17 and 18. The structure of these new compounds were elucidated by spectroscopic and chemical means. The occurrence of 8, 9, 11, 12 and 20 is of great interest from the viewpoint of biosynthesis of ipecac and Alangium alkaloids.
