New karyotypes of Crepis capillaris were artificially produced and established as stable culture lines. Tissue-cultured masses of shoot primordia were first treated with 5 μg/ml aphidicolin for 24 h, followed with 5 μg/ml mitomycin C (MMC) for 24 h. Cytochimerism of karyotypes, which was obviously observed upon termination of the MMC treatment, became gradually reduced during the recovery period. After 60 days of recovery, most of the shoot primordia showed karyotype homogeneity. However, some of the shoot primordia still contained a few variations in karyotypes. On day 120, completely homogeneous masses of shoot primordia with a mutated karyotype were found, each of which were isolated as culture lines. A variety of reciprocal translocations can be produced by this system.
