Perfusion pressure of pump-perfused hindquarters of the Wistar rats anesthetized with pentobarbital was measured. A dilator response was elicited by i.v. bolus injection of norepinephrine or by stimulation of the lumbar sympathetic nerves with pulses of 0.03 msec and shorter; longer pulses evoked a biphasic constrictor and dilator responses. The vasodilator responses were abolished by i.v. hexamethonium, indicative of their neurogenic nature. They were diminished by i.a. infusion of H1-antihistaminics, d, 1-chlorpheniramine and tripelennamine, but not by pyrilamine. However, d, 1-chlorpheniramine and tripelennamine, but not pyrilamine, are known to inhibit norepinephrine uptake, and were mimicked by cocaine in their antidilator activity. H2-antagonists, metiamide and cimetidine, given i.a. singly or in combination with pyrilamine had no such effect, not did i.a. atropine or propranolol. The latter two agents and H1- and H2-antagonists blocked the dilator responses to i.a. acetylcholine, isoprenaline and histamine, respectively. The neurogenic vasodilation was, therefore, probably not mediated by the H1- or H2-histaminic, muscarinic, or β-adrenergic receptors.
