The fine structure of neurosecretory cells of the rat hypothalamo-hypophyseal system was studied with the electron microscope. The role of cell organelles to the production of neurosecretory granules was discussed. Among many findings the direct continuity between the rough-surfaced endoplasmic reticulum and Golgi apparatus was noticed. Lysosomes were increased after repeated injection of trypan blue. Synapses on the cell surface were considered to be the site of neurohumoral reflex. Subsurface cisterns were observed beneath the synaptic contact. The neurosecretory granules did not alter their size during the transportation from the hypothalamic nuclei to the posterior pituitary. The neurosecretory substance may be released out in general by the mechanism of diacrine secretion. The limiting membrane of the secretory granule may become flat vesicles after the release of content and thereafter may be fragmented into many microvesicles, which have been referred to as synaptic vesicles.
