Separation of the Molecular Forms of Chicken Liver Cytosolic Aldehyde Reductase and Comparison with Mitochondrial Aldehyde Reductase(Biological)

The chicken liver cytosolic aldehyde reductases were resolved into six fractions (named F1-1, F1-2, F2-1, F2-2, F3 and F4) by ion-exchange and affinity chromatographies. The six fractions were each partially purified from a single homogenate of chicken liver and characterized. The enzymatic and physicochemical properties of cytosolic aldehyde reductases were compared with those of mitochondrial aldehyde reductases. All of the aldehyde reductases from both fractions had similar molecular weights of around 30000, and isoelectric points of 7-8. As judged from the substrate specificity, inhibitor sensitivity, kinetic studies and immunological analysis, mitochondrial aldehyde reductase I (M.AL.R.I) was distinct from all the cytosolic aldehyde reductases. Mitochondrial aldehyde reductase II (M.AL.R.II) was similar to F2-2.
