Kinetic Study on Cell Proliferation of Ehrlich Ascites Tumor Cells

Cytokinetic analysis was performed on the Ehrlich ascites tumor cells of hypotetraploid subline at the 4th and 7th day after the intraperitoneal inoculation of 1×10^6 cells, and the mean length of G_1, G_2 and S period was estimated by an autoradiographic method using simultaneous administration of Thymidine-6-^3H and Colcemid. The results obtained are as follows: mean length of G_1 period is 3 and 4.5 hours, and S period is about 8.5 hours and 9.0 hours, respectively. The mean duration of G2 period is about 1.5 hours for both ages of tumor cells. The dormant cell population is 16 and 25%, respectively. Assuming that the mean length of mitosis is one hour, generation time (i.g., sum of S+G_2+M+G_1) is 14 hours and 16 hours, respectively. From these results, it is concluded that the slowing of growth rate of Ehrich ascites tumor cells is brought about by an increase in the dormant cell population and lengthning of the G_1 period.
