In order to clarify the polyamin metabolism in the healing process of D-galactosamine-induced hepatitis the following results were obtained. (1) The activity of ornithine decarboxylase started to increase 14 hrs after administration of galactosamine aud reached 30 times the normal activity at about 25 hrs, the time of maximum severity of the hepatitis, as judged by levels of serum tlansaminases. (2) The content of putrescine increased in parallel with the activity of ornithine decarboxylase and reached about 10 times the control value. (3) Following the elevations in putrescine content and ornithine decarboxylase activity, the hepatitis was diminished. (4) Rise in activity of S-adenosylmethionine decarboxylase and in content of spermidine were observed 33-37 hrs after the administration of galactosamine. The maximum valuse of these parameters were significantly higher than those of controls and after that the healing process was observed. (5) Administration of putrescine to rats given galactosamine significantly decreased the severity of acute hepatitis, as judged by serum transaminases and hepatic glucose-6-phosphate dehydrogenase activities, and bλ histo pathological features. (6) Other diamines, such as 1, 3-diaminopropane and cadaverine, did not affect the levels in serum transaminases, but the administration of spermidine to rats reduced the high level of serum transaminases induced· by galactosamine. (7) A healing effect of putrescine was also observed in rats with thioacetamide-induced hepatic injury, as measured by change in their se measured by change in their serum transaminases but administration of spermidine to rats did not affect the levels in serum transaminases. These results indicate that a rise in tissue content of putrescine or its metabolites may be involved in the amelioration of hepatitis.
