Correlation between Electron-Dense Structures and Nuclear Inclusions Induced in Human Cytomegalovirus-Infected HEL Cells

Human cytomegalovirus (HCMV)-induced nuclear inclusions were examined electron microscopically after staining by the peroxidase-conjugated antibody/diaminobenzidine method and compared with HCMV-induced electron-dense, intranuclear structures which were stained with uranium and lead salts and detected by thin-section analysis. These results, together with those obtained with a temperature sensitive mutant of HCMV which produced electron-dense, internuclear structures but not nuclear inclusions at the nonpermissive temperature, suggested that the HCMV-induced nuclear inclusions are composed of virus-specific antigens accumulated around the skeleton of the electron-dense structure.
