Vascular endothelial growth factor(VEGF)has been shown to act as a maintenance factor as well as a growth stimulatory factor on sinusoidal endothelial cells in primary culture.In normal rat liver, VEGF and its receptors, VEGF-R1 and VEGF-R2, are expressed in hepatocytes and sinusoidal endothelial cells, respectively.When rats underwent 70% liver resection, VEGF expression was increased in hepatocytes later than 12h with a peak at 72h after the operation.In these rats, mitosis was maximal in hepatocytes at 36h and in sinusoidal endothelial cells at 96h, suggesting that VEGF derived from regenerating hepatocytes may be involved in reconstruction of the hepatic sinusoids through proliferation of sinusoidal endothelial cells.In contrast, in carbon tetrachloride-intoxicated rat liver, VEGF was markedly expressed in activated Kupffer cells, macrophages and stellate cells in the necrotic areas.These non-parenchymal cells seemed to induce capillarization of the hepatic sinusoids through proliferation of vascular endothelial cells, since they produced platelet derived growth factor(PDGF)and fibroblast growth factor(FGF)-2 as well.In these rats, reconstruction of the hepatic sinusoids occurred following absorption of the necrotic areas, when regenerating hepatocytes expressed VEGF abundantly.Recently, it is shown that VEGF-R1 expression was upregulated in activated stellate cells, and VEGF attenuated contraction of the cells in vitro.Also, VEGF increased the porosity of sinusoidal endothelial cells in primary culture on electron microscopy.Thus, a communication system between liver cells through VEGF may be involved in the regulation of hepatic microcirculation as well reconstruction and capillarization of the hepatic sinusoids.
