We have developed a culture system in which small hepatocytes(SHs)can grow forming colonies, when a nonparenchymal cell(NPC)fraction containing SHs and NPCs was cultured in the medium we devised(HCGM).To obtain the direct evidence that NPCs promote the growth of hepatocytes and to compare the growth potential between SHs and parenchymal hepatocytes(PHs), pure populations of SHs, PHs, and NPCs were prepared from the adult rat and co-cultures of hepatocytes and NPCs were reconstituted from them.SHs and PHs underwent multiple divisions, and the former showed a higher growth capacity than the latter when co-cultured with NPCs, whereas neither SHs nor PHs formed colonies when cultured alone.Stellate cells in the NPCs were demonstrated to be responsible for this growth promotion.We suggested that growth factors and extracellular matrices secreted by stellate cells as a result of cooperative interactions with hepatocytes stimulate the growth and differentiation of hepatocytes.
