16S rRNA is one of the essential components for protein synthesis in procaryotic cells. It has been known to contain species-specific nucleotide sequences which are able to identify bacterial species. The aim of this study is to clarify the diversity in the nucleotide sequences of 16S rRNA among Porphyromonos gingivalis strains. DNA fragments containing the 16S rRNA gene were amplified from genomic DNA samples of 5 strains of P. gingivalis (ATCC 33277,W83,FDC 381,SUNY 1021 and A7A1-28) by the PCR (polymerase chain reaction) method using primers specific to nucleotide sequences in the conserved regions of the 16S rRNA genes. Nucleotide sequences were analyzed by a dye terminating cycle sequencing reaction using the PCR amplified fragments as templates. The nucleotide sequences in the 28-30 regions of the helix were found to be identical among the 5 strains tested. The sequences seem to contain P. gingivalis species specific sequences. In region 37 of the helix, nucleotide substitution (U : G) was found at position 1006 in 1 of the 5 strains. These results suggest the availability of primer based on the 28-30 regions of the helix. This also suggests the specific amplification of the P. gingivalis 16S rRNA gene and possibility for the subclassification of P. gingivalis into two novel subtypes according to the nucleotide sequences in the 37 regions of the helix of 16S rRNA genes.
