Malignant fibrous histiocytoma (MFH), a common neoplasm among other soft tissue sarcomas, is known to show various histological findings mainly consisting of fibroblast-like and histiocyte-like cells. However, little has been known on its origin or detailed characteristics of the neoplastic cell. The element distribution in the cytoplasm and microorganella in unknown cells, was examined to provide some aspects on the metabolism of the neoplastic cell. Thus, the ultrastructural distribution of the essential elements contained in cultured MFH cells was examined by means of the electron probe X-ray microanalyzer which is a precision analytical tool for the quantitative localization of minute quantities of elements. In this study, specimens were obtained from cultured cells derived from a hamster MFH induced with DMBA in the cheek pouch, prepared by fixing with acrolein vapor, quick freezing, drying by replacement method with acetone, and embedding in ERL4206. After ultra-thin sections were made, they were mounted on Cu grids and observed using an electron microscope (Hitachi H-700H) equipped with an energy dispersive type X-ray micro-analyzer (Kevex K-7000Q). This procedure, enabled identification of the cytoplasm and micro-organella and elucidation of the ultrastructural element distribution in the nucleolus, nuclear envelope and cell membrane in the histiocyte-like cell obtained from the hamster's cheek pouch. The distribution of elements in respective micro-organella by weight retio was as follows : For the histiocyte-like cell, K>P>Na>S in the nucleolus, K>P>Na>S in the nuclear envelope, and P>Na>K>S in the cell membrane For the multinucleated giant cell, P>Na>K>S in the nucleolus, Na>P>S>K in the nuclear envelope, and Na>P>S>K in the cell membrane. For the normal cell from the hamster's cheek pouch, S>P>Na in the nucleolus, Na>S>P>K in the nuclear envelope, and Na>S>P>K in the cell membrane. In the analysis of energy intensity, all elements, Na, P, S and K were detected in the decreasing order of nucleolus, nuclear envelope and cell membrane in the histiocyte-like cell and multinucleated giant cell. In the normal cell from the hamster's cheek pouch, Na was detected in the decreasing order of nuclear envelope, cell membrane and nucleolus, P in the decreasing order of nuclear envelope, nucleolus and cell membrane S in the order of nucleolus, nuclear envelope and cell membrane, and K was detected in nuclear envelope as much as in the cell membrane, but not in the nucleolus.
