A ROLE OF DERMATOPONTIN IN WOUND HEALING

Dermatopontin (DPT)-null mice, generated by targeted disruption of Dpt gene, display abnormal collagen fibril formation and increased elasticity in skin. We have also shown the lack of DPT may cause decreased collagen deposition in skin. We think DPT-null mice could represent a useful animal model to investigate the wound healing, fibrosis, and tumorogenesis. Skin wound healing is a series of complex matrix-cell interactions that involve cellular migration and inflammation, followed by proliferation of fibroblasts with new collagen synthesis, and lastly tissue remodeling of the scar. To define the role of DPT during skin wound healing, mRNA expression levels and distributions were assessed by in situ hybridization. Samples were collected from the wild-type mouse wounds at 1, 4, 7,and 10 d after injury. Whereas very little expression was noted in early wounds (1 and 4 d), Dpt expression was upregulated at 7d after injury. Expressing cells were localized throughout the granulation tissue, and at no time did keratinocytes of the epidermis express Dpt. The pattern of Dpt expression was correlated with that of the collagen I expression. These findings suggest that DPT may function in remodeling wound matrix in a process that involves fibrillar collagen organization. To elucidate this hypothesis, we are currently studying the skin wound repair and scar formation by histological, ultrastrucural, and physiological analyses in DPT-null mice
