Tenascin was initially found to be an oncofetal type of extracellular matrix glycoprotein which is produced by the mesenchyme. The TN-gene family comprises the three distinct genes, TN / cytotactin(TN-C), J1-160 / 180 / janusin / restrictin(TN-R), and TN-MHC / XB / p450opp(TN-X). In mouse mammary glands, both immunoreactive TN-C and TN-C mRNA are demonstrated in the dense mesenchyme surrounding the embryonic mammary buds, growing end buds, and tumors. While in the epithelium, TN-C mRNA is expressed at the embryonic stage and tumors. In human breast cancers, TN-C is related to the prognosis. The positive staining in the cancer stroma is unfavorable and that in the cancer cells is much worse. For the biological functions of TN-C, we propose an anti-adhesion / counter adhesion and cancer cell growth promotion in collaboration with FN. In wound closure assay, TN-C stimulates the migration of the cancer cells cultured on the FN-coating plate. And in cancer cell growth assay, TN-C stimulates the proliferation of cancer cells cultured on FN-coating plates in dose-dependent manner.
