Morphological Regulation of Cultured Hepatic Stellate Cells by Extracellular Matrix through Intracellular Signaling

Hepatic stellate cells, which are located in the perisinusoidal space of Disse, extend their long processes around the sinusoid. In cell culture HSCs exhibit three different morphologies in response to the distinct substratum:(i)flattened shape with well developed stress fibers on the polystyrene surface, on the aminoalkylsilane- or lysine-coated surface, and on the type I or type IV collagen-coated surface, (ii)rod shape with well-developed cellular processes on or in type I or type III collagen gel, mimicking in vivo situation, (iii)round shape without a cell spreading and without process extension on or in the Matrigel containing the basement membrane components. The cellular processes are composed of microtubules as the core and F-actin at the periphery, and contain lipid droplets containing vitamin A and matrix metalloproteinase 1. The processes are induced by cell surface integrin-binding to interstitial collagen fibrils, followed by signaling processes including protein and / or phosphatidylinositol phosphorylation, and finally F-actin and microtubule organization.
