This study is aimed at clarifying the pathogenicity of Porphyromonas gingivalis (Pg) related to periodontal diseases, in terms of interactions between its lipopolysaccharide (LPS) and the subsets of splenic B cells. The cell cycle, cell proliferation, as well as the number of splenic B cell subsets were determined in examining cell alteration after PgLPS cultures were stimulated for 24h. Spleen cells were isolated from two mouse strains (C3H/HeJ and C3H/HeN) with different responses to PgLPS, while splenic B cells were purified from the cells by a negative selection method using an immunomagnetic column with proved to be an anti-non B cell monoclonal antibody. The purity of B cell preparations were 85% or more. The cell number after cultured for 24h culture decreased by half, of which 52〜63% was attributed to the decrease in naive B cells. There was about a 20% greater decrease due to the PgLPS stimulation of the B cells of the C3H/ HeJ mouse. These results suggest that PgLPS stimulation might be useful in delaying the start of the apoptosis of naive B cells as well as in its prevention.
