The purpose of this investigation was to examine vasomotor nerves following experimental tooth movemen in rat periodontal tissue pathologically, immunohistochemically and ultrastructurally. Orthodontic elastic was inserted between the crown of the first and second molars of the upper jaw, using Waldo's method. The animals were sacrificed 1,3,5,7 and 14 days after insertion of the elastic. Samples for pathological observation under a light microscope were fixed with neutral buffered Zamboni for 1 day. Sections were cut and stained with hematoxylin-eosin. For the immunohistochemical observation, samples were treated with antisera for the S-100 protein, and then treated with streptavidin-biotin peroxidase using a streptavidin-biotin kit. Some specimens were prepared for transmission electron microscopy. The results were as follows : 1. Hyaline degeneration of the periodontium and a decrease in nerve fiber and terminal axons around the arterioles in the pressure side were observed in the early stage after the beginning of tooth movement. However, alterations in the nerve fibers and terminal axons improved gradually during the later stage. 2. The nervous tissue in the pressure side was influenced significantly compared with that of the tension side. 3. In the pressure side, terminal axons with vasomotor nerve synaptic vesicles recovered. This recovery was accompanied by improvement in the circulatory disorder of the periodontium until 14 days after tooth movement. 4. It is suggested that the nerve ending of the vasomotor nerve with synaptic vesicles slightly regulated blood flow in the periodontium of the pressure and tension sides, beduese to very few synaptic vesicles in the nerve ending.
