The accumulation of CD4^+CD45RO^+ T cells has been found in inflammed periodontal tissues. Interleukin (IL)-17 is one of novel cytokines produced by only activated CD4^+CD45RO^+ T cells. The author aimed to examine the production of IL-17 mRNA in inflamed periodontal tissues and the effects of recombinant IL-17 (rIL-17) on proliferative activity and cytokine production of activated peripheral blood mononuclear cells (PBMCs) in order to know the association of IL-17 with progression of periodontal disease. The production of IL-17 mRNA was detected in the inflamed periodontal tissues from patients with adult periodontitis. The production of IL-17 mRNA seemed to be concomitant with those of IL-6,IL-8 and IL-16 mRNAs among subjects. These indicate that IL-17 might affect the production and secretion of IL-16 in periodontal lesions. The inhibitory effect of rIL-17 was found on proliferative activities of PBMCs activated by phytohemagglutinin. The effects of rIL-17 were found to be inhibitory on interferon-γ mRNA production of PBMCs activated with P. gingivalis whole cells in one out of two subjects, seemingly dependent on HLA class II genotypes. The role of IL-17 in periodontal disease is suggested to be (1) induction of memory Th2 predominant immune reactions by inhibition of the proliferation of naive T cells and production of type I cytokines in periodontal lesions and (2) recruiting immunocompetent cells to periodontal lesions by promoting production of various chemokines and IL-16 as well as IL-6 and IL-8.
