The effects of the upper incisor separation with three kinds of orthodontic strength on brain including cerebrum, cerebellum and brain stem, adrenals and three major salivary glands of SD rats were examined physiologically, biochemically and genetically during 7 days after treatment. The findings were as follows : 1. The wet weights of the submandibular (SMG) and sublingual (SLG) glands were significantly enlarged 3 and 7 days after treatment, but no effects was observed with increasing strength. 2. The norepinephrine (NE) and dopamine (DA) levels of brain significantly increased 7 days, whereas DA levels of adrenals significantly increased 1 and 7 days after treatment. NE and DA levels of three major salivary glands were significantly reduced 7 days in contrast that they significantly increased 1 and 3 days after treatment. Only NE levels of SMG significantly increased with increasing strength 7 days after treatment. 3. The salivary volumes in response to isoproterenol (IPR) had a tendency to be significantly reduced 3 and 7 days after treatment. The salivary volumes, the protein concentration and the amounts of protein secreted in response to IPR were reduced in rats subjected to stronger orthodontic appliances. 4. The amounts of two peptide peaks (P21 and P29) in IPR-stimulated saliva significantly increased 1 and 7 days, whereas those of three peaks (P2.4,P26 and P33) were significantly reduced 1,1 and 7 days after treatment, respectively. The amounts of two peaks (P2.4 and P27) were reduced with increasing strength. 5. The salivary cystatin S mRNA expression in the SMG increased 7 days after treatment by the RT-PCR method. It was conclusively suggested that with the upper incisor separation three major salivary glands, brain and adrenals of rats were affected with the organ-specificity, and especially SMG could be stimulated via β_1-receptors in the sympathetic nervous system. In addition, the effects of an orthodontic appliance was more prominent in changes of the duration length than in those of the degree of strength.
