It is very important to discriminate biomakers in saliva between patients with periodontitis and healthy men. Little information, however, is available on fluid and protein secretion by the submandibular glands (SMG) of plaque-susceptible (SUS) male and female rats with reference to salivary cystatin S (RSCS). The present study was thus designed to determine whether there were biomarkers in saliva different between SUS and plaque-resistant (RES) rats and to determine whether there was a stimulated system via the adrenal-sympathetic nerve in the RES and SUS rats. Submandibular saliva, extracts of SMG and gingiva, catecholamines of SMG and adrenal glands, and sera were analyzed by various methods including electrophoresis, spectrometry, ELISA, Ouchterlony, Western and Northern blotting. Amino acid sequence of RSCS purified was also analyzed by the Edman method. The results were as follows : 1. The reduced wet weights of SMG and sublingual glands of SUS rats were affected by the reduction of body weight. However, they responded more prominently to chronic treatment with isoproterenol (IPR). 2. The volumes of saliva secreted by SMG of SUS rats in response to various types of agonists tended to be reduced due to reduced wet weights of SMG and body weight in general. 3. Protein secretion by SMG of RES rats was stimulated in response to bioamines, but not other agonists. 4. The types of protein secreted by SMG following various agonists were not greatly different between both groups except a RSCS band, but responses of SMG following an α 2-adrenoceptor agonist were reduced in both groups. 5. Esteroprotease activities of submandibular saliva elicited by IPR were not greatly different between both groups, but were significantly different between both groups after chronic treatment with IPR. 6. The concentration of RSCS in RES, but not in SUS, rats increased significantly and its difference between both groups increased greatly with increasing ages. 7. The isoelectric point, molecular weight, some biochemical properties, antigenicity and amino acid sequence of N-terminal 26 amino acids of RSCS purified from RES rats were identical with those of RSCS purified from Sprague-Dawley rats. 8. The SMG of male and female SUS rats was more greatly affected than that of those RES rats by chronic treatment with IPR and no significant difference between both groups was seen in RSCS levels of SMG saliva as well as in extracts of SMG and gingiva, and sera after chronic treatment with IPR, although great differences between both groups were seen in these samples without any chronic treatment with IPR. 9. The expression of mRNA for RSCS increased dramatically in SMG of RES rats and Wistar rats subjected to chronic treatment with IPR. 10. The pocket depth of SUS rats became significantly deeper and deeper with increasing ages and after chronic treatment with IPR it was significantly shallower in SUS rats than that of SUS rats without any treatment with IPR. 11. The concentrations of adrenaline and noradrenaline, but not dopamine, in SMG and adrenal glands increased significantly in RES rats, when compared with those of SUS rats. From these results, it can be concluded that RSCS may become a biomarker between both groups and may probably be related to plaque-resistant. In addition, a stimulated system via the adrenal-sympathetic nerve could be involved in both rats.
