The objective of this study was to clarify the histological changes in taste buds and peripheral nerve of the Wistar rat lingual papillae. Sixty-two Wistar-strain male rats, six weeks old, were used in this experiment. To determine changes in nerve fibers in and around taste bud cells, immunohistochemical screening was carried out by using, Protein Gene Product (PGP), Substance P (SP) and Calcitonin Gene-Related Peptide (CGRP). The results obtained can be summarized as follows. Before parathyroidectomy, the serum Ca^<2+> concentration of normal rats used in this experiment was 9.99 mg/dl on average ; however, it began to fall three days after parathyroidectomy, reaching 7.26 mg/dl in one week, which was the lowest value. After that, the value gradually increased up to 7.99 mg/dl five weeks after parathyroidectomy. After parathyroidectomy, one week after administering a dosage of 0.1 μg/kg Vitamin D_3,normal values of the serum Ca^<2+> concentration were seen. Taste bud cells increased from the second day to the fifth week after parathyroidectomy, showing PAS positive granules in partial taste bud cells. These PAS positive granules were completely digested by α-amylase. There were few PAS positive granules in the Vitamin D_3 administration groups. As for parathyroidectomized groups, the decline of ATPase activity could be seen in some portions one week after parathyroidectomy. In five weeks, there was a significant decline in the number of taste buds located on the surface of the fissure lining a cleft. All of the taste bud cells in Vitamin D_3 adminstration groups returned to normal. Normally, SP positive fibers and only a few CGRP fibers existed in taste buds cells ; however, in this experiment they started to decline from the first week after parathyroidectomy and continued declining until the fifth week. Especially during the fifth week, they could only rarely be observed. In Vitamin D_3 administration groups, PGP positive fibers and positive cells were confirmed to be close to the normal distribution. In parathyroidectomized groups, SP, and CGRP positive fibers in lamina propria papilla started to increase from the third day, contacting the taste bud basal area, while in Vitamin D_3 administration groups those quantatives became normal. This experiment showed that the decline of serum Ca^<2+> by parathyroidectomy did not cause a sudden change or disappearance of taste buds cell, but indicated a decline of metabolism in the taste bud cells and the activity of the membranes. Furthermore, this study proved that there is a taste disorder development due to the decline of serum Ca^<2+>, and Vitamin D_3 administration enables taste bud cells to return to normal
