It has been elucidated that by chronic administration of a β-agonist, isoproterenol (IPR) , synthesis of cysteine proteinase inhibitors (cystatins) was dramatically induced in the submandibular glands of rats. This study was designed to determine the properties of mouse salivary cystatins and to elucidate whether the similar induction of their synthesis by IPR, as seen in rats, occurred. Two types of cystatins were purified from whole saliva of chronically IPR-treated (30 mg/kg body weight once a day for 7 consecutive days) female ddY mice by sequential Mono Q or Mono S ion exchange and CM-Papain Agarose affinity chromatographies. Mouse salivary acidic cystatin (MW. 12,900 and pI 4.6 determined by SDS-polyacrylamide gel and isoelectrofocusing electrophoresis) was designated as MSC-A and basic one (MW. 14,500 and pI 9.1) as MSC-B. Papain and ficin, cysteine proteinases from plants, were strongly inhibited with MSC-B but weakly with MSC-A. Amino acid composition of both MSCs was analyzed by the method of PTC amino acid analysis. Amino acid composition of MSC-B was very similar to that of mouse post γ-globulin described previously. N-terminal amino acid sequences of MSC-A (23 residues) and MSC-B (25 residues) were determined by the method of Edman and compared with other two cystatins. It was shown that N-terminal amino acid sequence of MSC-A was 30% identical to that of rat cystatin S and that of MSC-B was 43% identical to that of human cystatin C. Each of immunoreactive substances with anti MSC-A IgG and anti MSC-B IgG was localized in the acinar cells of the submandibular and parotid glands, but not in the sublingual glands, of control mice. Enlargement of mouse submandibular and parotid glands was induced by IPR chronic treatment, although the production of MSC-A and MSC-B was not markedly induced by IPR as occurs in rats. It was concluded that two types of cystatins were present in salivary glands of mice. Both cystatins, different from a rat cystatin S, were not markedly induced by IPR.
