Analysis of Miscanthus sacchariflorus and M. sinensis chromosomes by fluorescence in situ hybridization using rDNA and total genomic DNA probes

Chromosomes of Miscanthus sacchariflorus (2n=76) and M. sinensis (2n=38) were investigated by fluorescent in situ hybridization (FISH), using 18S-5.8S-26S and 5S rDNA probes to localize rRNA genes. Signals representing 18S-5.8S-26S rDNA were detected at the secondary constrictions of all four SAT chromosomes in M. sacchariflorus and at those of two SAT chromosomes in M. sinensis. FISH signals of 5S rDNA were detected at the interstitial regions of the four median chromosomes in M. sacchariflorus and at the interstitial regions of two median chromosomes in M. sinensis. The chromosomes of M. sacchariflorus were analyzed by genomic in situ hybridization (GISH) with a probe from M. sinensis total genomic DNA to investigate genome relationships between the two species. GISH signals appeared over all chromosome arms of M. sacchariflorus, with a pair of strong signals at each centromeric region. The results indicated that the genomes constituting tetraploid M. sacchariflorus and diploid M. sinensis could not be differentiated at the repetitive DNA level in their FISH karyotypes.
