FISH detection of sugar cane centromeric repetitive sequences in the chromosomes of two Miscanthus species

The chromosomal distribution of sugar cane centromeric repetitive sequences was examined by fluorescent in situ hybridization (FISH) in tetraploid Miscanthus sacchariflorus (2n=76) and diploid M. sinensis (2n=38). Using centromeric repetitive DNA pSG1-2 cloned from sugar cane as a probe, FISH signals appeared in the centromeric region of most Miscanthus chromosomes, except for several chromosomes from both species that had very weak or absent signals. When FISH were performed using probe DNAs amplified from genomic DNA of Saccharum officinarum, M. sinensis and M. sacchariflorus using SCEN family repetitive DNA specific primers, signals appeared at the centromeric regions of all chromosomes of M. sacchariflorus and M. sinensis. These FISH results demonstrated that M. sacchariflorus and M. sinensis had centromeric repetitive sequences that were highly homologous to the centromeric repetitive sequences of sugar cane. The results also suggested that the centromeric sequences within these genomes were more complex than the sequences included in the pSG1-2. The probes hybridized with different signal intensities to the centromeres of different chromosomes, suggesting that the centromeric regions of Miscanthus chromosomes displayed varying levels of homology. The sequence homology of these centromeric regions, as determined by FISH, coincided with the relatedness of the genera as deduced from taxonomic and phylogenetic analyses.
