1. The formation of buds and roots in seven strains of carrot root callus successively cultured for various periods on a medium containing WHITE'S inorganic salts, sucrose, 2, 4-D and yeast extract was investigated. 2, 4-D completely suppressed organ formation during stock subculturing. It was confirmed that the organ forming capacity of the callus in a 2, 4-D-free test medium diminishes and finally completely disappears with prolonged periods of previous subculturing of the callus. 2. IAA promoted the root formation. Yeast extract, casein hydrolysate and amino acids mixture promoted the bud formation of calluses at earliest steps of subculturing (Phase I). 3. At next steps of subculturing (Phase II). IAA-dependent root forming capacity of calluses was lost although the bud forming capacity induced by yeast extract, casein hydrolysate and amino acids mixture was retained. 4. At further advanced steps of subculturing (Phase III), yeast extract induced only root formation, while casein hydrolysate still could induce buds. IAA and amino acids mixture did not affect the organ formation. 5. No organ formation was observed in calluses subcultured over 38 months under any conditions attempted (Phase IV). 6. Single cells or small cell clumps obtained from the callus subcultured for 2 months formed only roots on the medium containing IAA and formed buds and roots on the medium containing yeast extract. 7. These differences in organ forming capacity and in responses towards various factors are interpreted to reflect the changes in physiological states of the callus during successive cultures on the stock culture medium.
