Purification and Characterization of Isolectins from Lycoris aurea

Two isolectins were obtained from bulbs of Lycoris aurea, Amaryllidaceae. Their properties were characterized and their carbohydrate-binding specificities were investigated by sugar-hapten inhibition of both precipitation and hemagglutination. The properties of the lectins were very similar to those of other lectins from the Amaryllidaceae. However, an antiserum raised against these lectins reacted weakly with other lectins from the Amaryllidaceae, revealing differences in immunological reactivity. The hemagglutination activity of the lectins were weakly inhibited by D-mannose and strongly inhibited by low concentrations of various glycoproteins, although hemagglutination of lectins from other members of the Amaryllidaceae reported to date is effectively inhibited by low concentrations of D-mannose (less than 20 mM) but only by high concentrations of thyroglobulin (over 1 mg ml^<-1>). Thus, the isolectins from Lycoris appear to be exceptional among lectins from the Amaryllidaceae.
