The addition of 3-O-methyl-D-glucopyranose to wild-type cells of Chlorella vulgaris (211-11 h) grown in glucose medium in the dark induced a rapid alkalization of the external medium (proton uptake), whereas this pH shift did not occur in autotrophically grown cells. Light-irradiation inhibited the sugar-induced proton uptake, making the blue end of the visible spectrum very effective and the red end only slightly effective. This spectral dependence corresponds to that of photoinhibition on hexose uptake in these algae. A similar photoinhibitory effect was observed in cells of a colorless mutant of Chlorella vulgaris (M125). The activity of nitrate-proton symport in Chlorella vulgaris (211-11 h) was also enhanced by the addition of glucose. Illumination had no inhibitory effect on this increased transport. The effect of light on the hexose uptake system is discussed.
