Changes in Lipid and Fatty Acid Metabolism of Vicia faba Mesophyll Protoplasts

The metabolism of the major polar and neutral lipids of Viciafaba protoplasts isolated from ^<14>CO_2-fed leaves has been examined. The results show large losses in the radio-activity found in phosphatidylcholine and monogalactosyldiacylglycerol while that of phosphatidylglycerol was stable. This loss was accounted for by a rapid increase in the ^<14>C content of the neutral lipids, particularly the triacylglycerols. Analysis of the fatty acid radioactivity in the lipids suggests that protoplast isolation inhibited fatty acid desaturation on phosphatidylcholine and possibly on other lipids. These results also suggest a role of phosphatidylcholine in the donation of fatty acids for triacylglycerol synthesis in mesophyll protoplasts. The results are discussed in terms of the regulation of lipid metabolism and protoplast biology.
