Two Different Soluble Cytoplasmic Auxin-Binding Sites in Etiolated Pea Epicotyls

Etiolated pea epicotyls from 7-9 day-old seedlings exhibit the existence of only one class of soluble auxin binding sites, while later stages (10-12 days) obviously have two: A second class of sites appears in a protein fraction which by means of chromatofocusing can be separated from the first. The pI-ranges are 5.0-6.0 for the first and 6.0-6.8 for the second class of sites, respectively. Double reciprocal plots show that there is no competition between the sites, which obviously have different K_D (0.55 × 10^<-7> M vs. 2.24 × 10^<-7> M). Evidence is presented that soluble auxin binding is not likely to be due to binding to NAA-conjugating enzymes, since no conjugation products can be found in supernatants after binding assays.
